      目的 探究冬凌草对高糖-高脂诱导的非酒精性脂肪性肝病大鼠模型线粒体功能及氧化应激的影响。方法 选取60只雄性SD大鼠,采用随机数字表法分为对照组、模型组、低剂量冬凌草组(4 g·kg-1·d-1)和高剂量冬凌草组(8 g·kg-1·d-1),每组15只。除对照组外各组大鼠采用高脂饲料喂养制成非酒精性脂肪性肝病大鼠模型,低剂量冬凌草组和高剂量冬凌草组分别使用4 g·kg-1·d-1和8 g·kg-1·d-1的冬凌草灌胃治疗3周,对照组和模型组采用等剂量的0.9%氯化钠注射溶液灌胃。测定大鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸转氨酶(AST)、r-谷氨酸转移酶(GGT)含量水平;检测大鼠肝脏线粒体含量;测定细胞色素C氧化酶活性;检测线粒体膜电位及血清氧化应激标志物丙二醛(MDA)、超氧化物歧化酶(SOD)、乳酸脱氢酶(LDH)的含量水平。结果 相比对照组,模型组大鼠体质量、ALT、AST、GGT、MDA、LDH含量水平显著升高,线粒体含量、细胞色素C氧化酶活性、线粒体膜电位差、SOD含量水平显著降低(P<0.05);相比模型组,使用冬凌草处理各组大鼠体质量、ALT、AST、GGT、MDA、LDH含量水平显著降低,且高剂量冬凌草组大鼠显著低于低剂量冬凌草组(P<0.05);相比模型组,使用冬凌草处理各组大鼠线粒体含量、细胞色素C氧化酶活性、线粒体膜电位差、SOD含量水平显著升高,高剂量冬凌草组大鼠显著高于低剂量冬凌草组(P<0.05)。结论 冬凌草通过保护线粒体功能并抑制氧化应激反应治疗非酒精性脂肪性肝病,在一定浓度范围内呈剂量依赖性。

      Objective To explore effects of Rabdosia rubescens(RR) on mitochondrial function and oxidative stress in rat model of nonalcoholic fatty liver disease (NAFLD) induced by high-glucose and high-fat.Methods Sixty male SD rats were selected and divided into control group,model group,low-dose RR group (4 g·kg-1·d-1) and high-dose RR group (8 g·kg-1·d-1) according to the random number table method,15 cases in each group.In addition to the control group,non-alcoholic fatty liver disease rats were fed with high-fat diet.The low-dose RR group and high-dose RR group were intragastrically administered with 4 g·kg-1·d-1 and 8 g·kg-1·d-1 RR for 3 weeks,respectively.The control group and model group were intragastrically administered with an equal dose of normal saline.The levels of serum alanine aminotransferase (ALT),aspartate aminotransferase (AST) and r-glutamate transferase (GGT) were measured by automatic biochemical analyzer.The contents of mitochondriainrat liver,the activity of cytochrome C oxidase,the mitochondrial membrane potential (MMP),the levels of oxidative stress markers such asmalondialdehyde (MDA),superoxide dismutase (SOD) and lactate dehydrogenase (LDH) were detected.ResultsCompared with those in the control group,the weight,ALT,AST,GGT,MDA and LDH were significantly increased,and mitochondria content,activity of cytochrome C oxidase,MMP difference and SOD content were lowered in model group (P<0.05).Compared with those in the model group,the weight,ALT,AST,GGT,MDA and LDH were significantly decreased in groups treated with RR,and the above indexes in high-doseRR group were significantly lower than those in low-dose RR group (P<0.05).Compared with those in the model group,the mitochondria content,activity of cytochrome C oxidase,MMP difference and SOD content were significantly increased in groups treated with RR,and the above indexes in high-dose RR group were significantly higher than those in low-dose RR group(P<0.05).Conclusion RR can treat NAFLD by protecting mitochondrial function,inhibiting oxidative stress reactions and showing dose-dependence within certain concentration range.
